Following on from her Golden Globe best actress win last month, US star Moore picked up the same award for her performance in horror The Substance on Friday at the Barker Hangar in Santa Monica.
The ceremony saw The Substance, Emilia Perez and Wicked each win three trophies, while Japanese drama Shogun, comedy series Hacks and Netflix hit Baby Reindeer picked up major awards in the TV categories.
Collecting her award, Moore, 62, said: “This has been such a wild ride, I can’t tell you when I started this journey on this film, I could never imagine being here.
“This is so far beyond anything I could have hoped for.”
The actress added: “For anybody out there who is still on their journey, who is still struggling to find their way… because it hasn’t happened, it doesn’t mean it isn’t happening, dreams do come true.”
The film, which sees Moore’s character confront the harsh reality of ageing in Hollywood, also won best original screenplay for filmmaker Coralie Fargeat and best hair and makeup.
Anora, a film about a young sex worker who marries the son of a Russian oligarch, took home the best picture prize at the end of night after missing out in earlier categories.
Wicked won best director for Jon M Chu, who helmed the film adaptation of the hit Broadway and West End musical starring Ariana Grande and Cynthia Erivo, as well as best production design and best costume design.
Best supporting actress was awarded to Zoe Saldana for her role in Jacques Audiard’s musical Emilia Perez, a thriller about a Mexican drug lord who changes gender.
It also took home best foreign language film and best song for El Mal which features Saldana, Karla Sofía Gascon and Camille Dalmais.
Elsewhere, Adrien Brody won best actor for his starring role as a Hungarian-Jewish architect in The Brutalist, following on from his Golden Globe in the category last month.
Deadpool & Wolverine, starring Hollywood actors Ryan Reynolds and Hugh Jackman, won the best comedy prize.
Japanese drama Shogun dominated in the TV categories, securing best drama, best actor for Hiroyuki Sanada, best supporting actress for Moeka Hoshi and best supporting actor for Tadanobu Asano.
Hacks picked up best comedy series and best actress in a comedy series for Jean Smart, while Adam Brody secured the best actor in a comedy series gong for his turn in Netflix hit Nobody Wants This.
